- 1) Do not eat, drink, smoke or chew gum for 30 minutes before collecting samples.
- 2) The swab tips should not come into contact with any surface other that the inside of the cheek at any point before, during or after collection
- 3) Move the swab up and down while also rotating. It's very important to rotate the swab with your fingers.
- 4) Allow the swab to dry before storing. At least one hour.

How do I read my DNA Sibling Test results report?
Brief introduction
In this article we will present some guidelines that must be taken into account when knowing how to interpret results about DNA tests between alleged siblings or half-siblings.
To be able to identify the results, you need to know a little about it, and to take into account some scientific data, which are not difficult to understand.
Many times, we receive the preliminary report - the results of a DNA test - and we do not know how to recognize the results of it. So, here we bring you a little information about this genetic test, and so that you know how to read the solution of the study that you and your brother have successfully carried out.

Data to interpret a DNA test between siblings
In order to better understand the details of the sibling test results, it is important to emphasize that you are familiar with the terms for proper understanding, and the science behind how a sibling DNA test works.
In this test, our laboratory generates a DNA profile with specific genetic markers (also known as short tandem repeats or STRs) at a set of known locations (loci or singular, locus) in your DNA.
At each DNA locus, you can find 2 genetic variants - known as alleles - which are each inherited, one by the mother and one by the father.3
The laboratory technique that is used to amplify the DNA study is called polymerase chain reaction (PCR). In a sibling DNA test, PCR analysis is used to determine which alleles are specific, which ones they have at each DNA locus, also known as the genetic profile. If two individuals share at least one allele in common, it means that a match already exists.
True siblings or half-siblings need to match on some alleles. This is because siblings don't always inherit the same DNA markers from their parents; Therefore, if this were the opposite case, all the brothers would be practically identical.
Interpreting your results
As soon as you have the report of the results in your hands, here you will find a graph that represents the DNA profiles of each person who has participated in the analysis.

The numbers in the Putative Sibling 1 and Putative Sibling 2 columns are values that represent the two alleles that you and your putative sibling received from your parents at the corresponding genetic marker. You can have two identical alleles on a marker (for example the number 14), or you can have two different alleles (for example the numbers 13,14). If the same number is present in both columns, that means you match the corresponding genetic marker of your suspected sibling.
DNA Locus | Putative Sibling One | Putative Sibling One | Sibling Index (Full) |
Sibling Index (Half) |
D8S1145 | 13,14 | 14 | 1.32 | 1.57 |
D24S54 | 28,30 | 28,30 | 4.25 | 1.72 |
The first column, labeled DNA Locus, lists those genetic markers that have been tested for that sibling DNA test (genetic marker example: D8S1145).
The final two columns, both Sib Rate (Full) and Sib Rate (Half), represent the statistical strength of a single match at the corresponding genetic marker. These values are calculated based on the type of match, which is normally expected as positive results for true pairs, between full siblings, and both half siblings.
Guild Composite Index
Sibship Index | Explanation |
0.01 | The individuals are 100% more likely not to be true siblings than siblings. |
10 | The individuals are 10% more likely to be true siblings than not siblings. |
100 | The individuals are 100% more likely to be true siblings than not siblings. |
A higher sibling index (greater than 1.0) corresponds to a higher probability of kinship. The higher that value is above 1.0, the greater the probability that the two individuals are related as true siblings.
Order your brotherhood test now and get rid of your doubts about whether your brother is a full brother, a half brother, or if he does not have any type of genetic relationship: Order now!
If, on the other hand, a brotherhood index (less than 1.0) corresponds to a lower probability of kinship. The lower this sibling index is less than 1.0, the lower the probability that the two individuals are related as identical blood siblings.
So how do we know if we are half siblings or full siblings?
The division that exists to calculate the percentage of brotherhood that exists between two presumed brothers is the following:
You must divide the Combined Sibship Index divided (full) by the Combined Sibship Index (half).
In such a way, based on an example: if a pair of brothers has a complete brotherhood index of 1.2 and an average brotherhood index of 40, it means that both are 33.3 times more likely to be half brothers than full brothers. (in numeric text mode this is 40 ÷ 1.2 = 33.33 percent chance).
This is how you can calculate and interpret your sibling DNA test results.
